Research details

Areas of research interest

PT-141 is studied for sexual desire and arousal, acting on brain pathways rather than on blood flow. Bremelanotide is the active ingredient in an FDA-approved prescription medicine for low sexual desire in premenopausal women. This research-grade material is not that product and is not for human use.

Overview

PT-141 (bremelanotide) is a synthetic peptide analogue of alpha-melanocyte-stimulating hormone (α-MSH). It activates melanocortin receptors in the brain, mainly MC4R in the hypothalamus. This central mechanism differs from PDE5 inhibitors, which act on blood flow.

Research summary

Phase 3 trials. The two RECONNECT trials randomized about 1,250 premenopausal women with hypoactive sexual desire disorder to bremelanotide or placebo [1]. Bremelanotide produced statistically significant improvements in sexual desire and reductions in desire-related distress at 24 weeks [1].

Long-term extension. In a 52-week open-label extension, no new safety signals emerged, and improvements in symptoms were sustained [2].

Regulatory status. FDA approved bremelanotide in June 2019. Prescribing information advises against use in people with cardiovascular disease or uncontrolled high blood pressure because of transient blood pressure increases.
