Areas of research interest
Melanotan-II has been studied for skin pigmentation and, through its action on brain melanocortin receptors, for sexual function. That second line of research led to the development of bremelanotide (PT-141). Melanotan-II has no approved indication anywhere in the world.
Overview
Melanotan-II is a synthetic cyclic analogue of alpha-melanocyte-stimulating hormone. Unlike Melanotan-I, it is not selective: it activates several melanocortin receptors (MC1R, MC3R, MC4R, and MC5R), which links pigmentation effects with effects on appetite and sexual function.
Research summary
Early clinical studies. A pilot Phase 1 study in the 1990s evaluated its pigmentation effects [1], and a small study in men with psychogenic erectile dysfunction reported that it initiated erections [2]. No completed efficacy trials have led to approval.
Safety signals. Published case reports have linked unregulated Melanotan-II use to melanoma, changes in moles, priapism, rhabdomyolysis, and kidney infarction. These are individual reports, not controlled data, but they are a consistent theme in the literature.
