Research details
Areas of research interest
Tirzepatide is studied mainly for glycemic control in type 2 diabetes and for body-weight reduction. Because it acts on both GIP and GLP-1 receptors, it serves as a common reference compound in incretin research. Tirzepatide is the active ingredient in FDA-approved prescription medicines. This research-grade material is not one of those products and is not for human use.
Overview
Tirzepatide is a synthetic 39-amino-acid peptide that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. A C20 fatty diacid side chain promotes albumin binding and extends its circulating half-life.
Research summary
Type 2 diabetes. In the SURPASS-2 trial of 1,879 adults with type 2 diabetes, all three tirzepatide arms were noninferior and superior to semaglutide for HbA1c reduction at 40 weeks, with greater body-weight reduction [1]. Gastrointestinal events were the most common adverse effects in both groups [1].
Obesity. In SURMOUNT-1, 2,539 adults with obesity or overweight but without diabetes lost a mean 15.0% to 20.9% of body weight at 72 weeks across the three tirzepatide arms, compared with 3.1% on placebo [2]. Adverse events led to discontinuation in 4.3% to 7.1% of tirzepatide participants [2].
Regulatory status. FDA approved tirzepatide for type 2 diabetes in May 2022. Later approvals covered chronic weight management (November 2023) and obstructive sleep apnea in adults with obesity (December 2024).
