Research details

Areas of research interest

Semaglutide is studied for glycemic control in type 2 diabetes, body-weight reduction, and cardiovascular risk. It is the most widely used reference GLP-1 receptor agonist in current metabolic research. Semaglutide is the active ingredient in FDA-approved prescription medicines. This research-grade material is not one of those products and is not for human use.

Overview

Semaglutide is a synthetic GLP-1 receptor agonist built on the native GLP-1 sequence. An amino acid substitution at position 8 protects it from DPP-4 breakdown, and a C18 fatty diacid side chain promotes albumin binding for a prolonged half-life.

Research summary

Obesity. In STEP 1, 1,961 adults with obesity or overweight but without diabetes lost a mean 14.9% of body weight at 68 weeks, compared with 2.4% on placebo [1]. Nausea and diarrhea were the most common adverse events and were typically transient [1].

Cardiovascular outcomes. SELECT enrolled 17,604 adults with established cardiovascular disease and overweight or obesity but no diabetes [2]. Semaglutide was superior to placebo in reducing cardiovascular death, nonfatal heart attack, or nonfatal stroke over a mean follow-up of 39.8 months [2].

Regulatory status. FDA approved semaglutide for type 2 diabetes in 2017 and for chronic weight management in 2021. A cardiovascular risk-reduction indication followed in 2024.
