Research details
Areas of research interest
AOD-9604 was developed to study fat metabolism, aiming to keep growth hormone's fat-reducing activity without its effects on growth or IGF-1. Smaller research interest covers cartilage repair. Its obesity drug program ended in 2007 after a Phase 2b trial missed its weight-loss endpoint. AOD-9604 is not approved by FDA for any use.
Overview
AOD-9604 is a 16-amino-acid fragment of the C-terminal end of human growth hormone (residues 177 to 191 plus a tyrosine). This region of growth hormone carries much of its fat-metabolism activity.
Research summary
Animal models. In obese mice, chronic AOD-9604 treatment increased fat oxidation and reduced body weight [1]. The weight effect was absent in mice lacking the beta-3 adrenergic receptor, which points to that receptor in the mechanism [1].
Human trials. Across six randomized, placebo-controlled trials, AOD-9604 had no effect on serum IGF-1, and its safety and tolerability were indistinguishable from placebo [2]. However, a 24-week Phase 2b obesity trial in more than 500 adults did not achieve significant weight loss compared with placebo, and development was stopped [2].
